Protein Degradation by the 26S Proteasomes

In the ubiquitin (Ub)/proteasome proteolytic pathway, the substrate-linked polyUb chain is the specific targeting signal recognized by the 26S proteasome. However, the mechanisms by which the 26S proteasome recognizes, unfolds and translocates the protein leading to proteolysis remain poorly understood. In my laboratory, biochemical and genetic analyses will be used to address how such key functions are partitioned among different subunits of the 19S, focusing on ATP utilization of these actions. We are also interested in understanding how 26S proteasome can be inactivated. The significance of proteasome function was recently validated by the recent FDA approval of the use of proteasome inhibitor as a treatment for multiple myeloma.
